Clarification

Clarification is the physical separation of insoluble particles from a liquid phase in bioprocessing. Clarification in Downstream Bioprocess Engineering refers to the removal of suspended solids (i.e. cells debris, or large particulates) from liquid streams to produce a clarified fluid suitable for further purification. It is a critical early step that ensures downstream operations—such as chromatography —run efficiently and without fouling. Clarification techniques are used to reduce turbidity, protect downstream equipment, and prepare the product stream for high-resolution purification. Clarification relies on differences in particle size, density, and solubility to achieve separation.

Common Clarification Methods

  • Filtration: Uses porous media (depth filters, membrane filters) to trap particulates.

  • Settling (Sedimentation): Relies on gravity to separate heavier particles from liquid. Critical in wastewater treatment processes.

  • Centrifugation: Applies centrifugal force to separate particles by density.
